ABSTRACT

In order to obtain the natural products synthesized by the three putative xiamycin biosynthesis gene clusters which were predicted via antiSMASH during the genome mining of marine Streptomyces sp. FXJ 7.388, Streptomyces sp. FXJ 8.012, and Streptomyces olivaceus FXJ 7.023. Sixteen genes involved in xiamycin assembly, modification, and regulation with higher identity than the newest reported xiamycin biosynthetic gene cluster from marine Streptomyces sp. SCSIO 02999, Streptomyces sp. HKI0576, and Streptomyces sp. FXJ 7.388 were discovered via gene cluster comparative analysis. A ribosome engineering strategy was adopted to activate such cryptic gene clusters with different final concentrations antibiotics that act on the ribosome, and two indolosesquiterpenes were isolated from idlethaldose streptomycin-resistant Streptomyces sp. FXJ 7.388 strains. However, no such product was detected in Streptomyces sp. FXJ 8.012 and Streptomyces olivaceus FXJ 7.023 under the same treatment. This result suggested that these genes might hold the least gene content for xiamycin biosynthesis

ABSTRACT

IL-17 Receptor D (IL-17 RD) is a cytokine receptor that mediates IL-17 signaling and plays an important role in responding to the invasion of extracellular pathogens and many inflammatory and autoimmune diseases such as rheumatoid arthritis. In this study we report the generation of a mouse monoclonal antibody against human IL-17 RD. The recombinant human IL-17RD extracellular domain (hIL-17RD-ECD) was produced in the baculovirus expression system and purified from culture medium of sf9 insect cells. The purified protein was used as a T-dependent antigen to immune Balb/C mice. B cells from the spleen of immunized mice were fused with murine cell SP2/0. Hybridoma cell lines were screened for the production of the monoclonal antibody against hIL-17-RD-ECD using ELISA. A hybridoma cell line 1F8 was found to have a high production of the antibody, which was further confirmed for the specificity by both western blot and ELISA analyses. The monoclonal antibody obtained from hybridoma 1F8 was characterized to be IgG1+Kappa subclass. This study provided a base for the further therapeutic application of the antibody on the autoimmune disease including rheumatoid arthritis.

ABSTRACT

Sef (similar expression to fgf genes) was identified as a feedback antagonist of FGF signaling in zerbrafish, mouse and human. Sefhas been reported to function in different ways, however the regulation of Sef stability remains unknown. The possible role of c-Cbl in the regulation of Sef protein degradation was investigated. Results from coimmunoprecipitation and immunostaining assays reveal that hSef colocalizes and interacts with c-Cbl. Data suggest that the interaction between hSef and c-Cbl results in the ubiquitination and subsequent degradation of the hSef protein. It was proposed that c-Cbl may serve as a modulator to regulate Sef protein stability during FGF signal transduction.

ABSTRACT

Novel Oncogene with Kinase-domain (NOK) is a novel tumor-related gene, coding receptor like protein with a kinase domain. Overexpression of NOK leads to tumorigenesis and metastasis. To further study NOK function in physiological condition, it is necessary to prepare the anti-NOK antibody. In this report, GST fusion protein was adopted to prepare polyclonal antibodies against hNOK. The result showed that the antibodies we generated is with a very high titriation, and can be used for examination of NOK protein by Westernblot. Furthermore, the antibodies were used for immunohistochemistry in lung cancer tissues, and the results demonstrated high expression of hNOK in the tumor tissues. The antibody of hNOK we generated can serve as a diagnostic method for the lung cancer.

ABSTRACT

OBJECTIVE To find out the situation of bacteriological contamination of oxygen humidified fluid in the hyperbaric oxygen chamber in Nanfang Hospital and take appropriate measures accordingly to provide valuable reference for avoiding possibly resulting hospital infection.METHODS Before and after beginning periodically changing oxygen humidified fluid, the humidifying fluid in the oxygen humidifiers of three hyperbaric oxygen chambers in this hospital was sampled once a season. The samples were analyzed with bacteria counting and identification.RESULTS The average amount of bacteria in the samples before and after changing oxygen humidified fluid was(7.9 ? 104) CFU/ml and(2.8 ? 103) CFU/ml,respectively.Identified bacteria were conditioned pathogens, including Chryseobacterium,Acinetobacter baumannii,Escherichia coli,Pseudomonas aeruginosa,Candida albicans, etc.Among them the frequency of Chryseobacterium was the highest. The results had no correlation with the changing of seasons.CONCLUSIONS Hyperbaric oxygen chamber should be disinfected periodically, the humidified fluid should be changed every day,and the amount of bacteria in it should lower than 103 CFU/ml.

ABSTRACT

OBJECTIVE To analyze the high risk factors and etiology character of ventilator-associated pneumonia(VAP) and provide the guidance for preventing VAP. METHODS Eighty three cases given mechanic ventilation over 48 hours in ICU of our hospital from Jan to Oct 2005 were observed.Thirty seven cases met the diagnosis criteria of VAP,while 46 cases without VAP were taken as control. RESULTS Airway secretion was cultivated and Gram-negative germ was the majority(58.0%).The first three pathogens were Pseudomonas aeruginosa(32.0%),Acinetobacter,and Staphylococcus epidermidis,22.0% cases had mixed infection with fungi.Except antiacid treatment, the following factors, such as long time for mechanical ventilation, trachea incision, corticosteroids treatment and coma state had significant relationship with the occurrence of VAP. CONCLUSIONS The occurrence of VAP is related with multiple clinical factors.Clinicians should reduce high risk factors to lower the occurrence of VAP.

ABSTRACT

OBJECTIVE To study the characteristics of pathogens and risk factors of ventilator-associated pneumonia(VAP) in our hospital to determine the prevention and treatment strategies. METHODS Prospective observation was performed in all the ventilator-related patients in 2004. RESULTS Seventy eight events of ventilator-related pneumonia among 56 cases occurred from all the 202 ventilator-related patients, the infection incidence was 98/1000VDs.The revealed important risk factors were oldness, coma, tracheotomy, use of H-2 receptor inhibitors, and prolonged mechanical ventilation.64.8% of the pathogens were G-bacilli,among which Pseudomonas aeruginosa contributed majority accounting for 22.7%,fungi accounting for 20.5%.There was wide resistance to antibiotics in G+ pathogens, including all of the isolated Staphylococcus. CONCLUSIONS VAP is caused predominantly by G-bacteria,fungi,and multiple predisposing factors. The combined treatment is necessary for control of VAP.
